Will this make a difference if I already play in 4K?

Texture resolution and screen/game resolution are completely separate and independent of each other. Your screen and game are running in 4k but doesn't affect the textures, only the texture quality setting does. Not sure the standard res for Arthur's face on ultra but I'd imagine 2k. Texture resolution differences are easier to distinguish on higher resolution screens though so it would actually make more of a difference on a 4k screen, not less.

Honestly, I tried it, I'm not sure if my eyes deceive me but I couldn't spot the difference. I also play on DLSS Quality with Nvidia sharpening+. I also have textures on ultra, so I know I'm good on that front.

Maybe I'll inspect the difference more closely next time.

You should take a screenshot of both. That way you can switch the screenshots back and forth to notice the difference.

I've been using Arthur Redone textures on this playthrough. When I remove those textures and replace them with these the game gets frozen on load screen. Do i need to start a new playthrough to use these or is there something I can do to swap these mid playthrough?

The map is pretty high quality, and i don't know if this is a bug, or just that the skin tone and shades were changed, but arthur is very pale with this mod running. Screeshot of Arthur with the 4k facemap from this mod.

Mod causing a crash while loading into singleplayer... (no other mods in use)
